Controlling the controls: GAD65 autoreactive T cells in type 1 diabetes.
نویسنده
چکیده
The clinical onset of autoimmune (type 1) diabetes is associated with multiple immune abnormalities. First, the islets of Langerhans are infiltrated with macrophages and CD4 and CD8 T lymphocytes. This islet inflammation, or insulitis, is related to a major loss of islet β cells. In addition, the onset is strongly associated with autoantibodies against specific islet cell autoantigens. In particular, autoantibodies against the smaller isoform of glutamic acid decarboxylase (GAD65), the islet antigen-2 (IA-2, also known as ICA512), or insulin are common at the day of clinical onset. The simultaneous presence of all three autoantibodies is highly predictive of type 1 diabetes (1).

Detection of GAD65 Autoreactive T-Cells by HLA Class I Tetramers in Type 1 Diabetic Patients
Type 1 diabetes (T1D) is an autoimmune disease, in which pancreatic beta cells are destroyed in genetically predisposed individuals. While the direct contribution of autoantibodies to the disease pathogenesis is controversial, it is generally recognised that the mechanism of beta cell destruction is mediated by autoreactive T cells that had escaped the thymic selection. We aimed to design a met...
متن کاملEvidence for in vivo primed and expanded autoreactive T cells as a specific feature of patients with type 1 diabetes.
Identifying beta cell autoantigen-reactive T cells that are involved in the pathogenesis of type 1 diabetes has been troublesome for many laboratories. Disease-relevant autoreactive T cells should be in vivo Ag experienced. The aim of this study was to test this hypothesis and then use this principle as a strategy for identifying diabetes-relevant autoreactive T cells. In this study, a CSFE dil...
متن کاملGAD65-reactive T cells are activated in patients with autoimmune type 1a diabetes.
Insulin-dependent type 1 diabetes is an autoimmune disease mediated by T lymphocytes recognizing pancreatic islet cell antigens. Glutamic acid decarboxylase 65 (GAD65) appears to be an important autoantigen in the disease. However, T cells from both patients with type 1 diabetes and healthy subjects vigorously proliferate in response to GAD65 stimulation ex vivo, leading us to postulate that th...
متن کاملGAD65-specific CD4+ T-cells with high antigen avidity are prevalent in peripheral blood of patients with type 1 diabetes.
Negative selection of self-reactive T-cells during thymic development, along with activation-induced cell death in peripheral lymphocytes, is designed to limit the expansion and persistence of autoreactive T-cells. Autoreactive T-cells are nevertheless present, both in patients with type 1 diabetes and in at-risk subjects. By using MHC class II tetramers to probe the T-cell receptor (TcR) speci...
متن کاملIncreased Expression of TRAIL and Its Receptors on Peripheral T-Cells in Type 1 Diabetic Patients
Background: Type-I diabetes is an autoimmune inflammatory disease in which pancreatic ß-cells are selectively destroyed by infiltrating cells. TNF-related apoptosis-inducing ligand (TRAIL) is a type-II membrane protein of the TNF superfamily which is expressed in different tissues, including pancreas and lymphocytes. In humans, TRAIL interacts with four membrane receptors. TRAIL-R1 and TRAIL-R2...
